ABSTRACT

Objective·To investigate the role of mitochondrial solute carrier family 25 member 13 (SLC25A13) on breast cancer development.Methods·SLC25A13 mRNA and protein expressions in invasive breast cancer tissues and normal breast tissues were from The Cancer Genome Atlas (TCGA) breast cancer dataset. Survival analysis was conducted online by Kaplan-Meier software. MCF-7 cell line was used for in vitro cell assay.Knockdown of SLC25A13 and sirtuin 2 (SIRT2) were conducted by siRNA transfection. Cell viability was measured with trypan blue exclusion. Cell cycle arrest was determined by flow cytometry. The mRNA expression of SLC25A13 and P27 were detected by quantitative PCR. The protein level of SLC25A13, P27 and SIRT2 were detected by Western blotting. Protein half-life of P27 was assessed by Western blotting after ycloheximide treatment. Results·SLC25A13 was up-regulated in invasive breast cancer tissues. High expression of SLC25A13 correlated with poor overall survival and breast cancer recurrence. SLC25A13 knockdown inhibited MCF-7 cell cycle progression. P27 and SIRT2 both accumulated after SLC25A13 knockdown. P27 accumulation resulted from prolonged protein half-life. Knockdown of SIRT2 restored cell cycle arrest as well as P27 accumulation caused by SLC25A13 silencing. Conclusion·High expression of SLC25A13 may promote cell cycle progression via SIRT2 in breast cancer development.

ABSTRACT

<p><b>Background</b>Paclitaxel (PTX) could inhibit the growth of fibroblasts, which occurs in proliferative cholangitis and leads to biliary stricture. However, its use has been limited due to poor bioavailability and local administration for short time. This study designed and synthesized a new PTX-conjugated chitosan film (N-succinyl-hydroxyethyl chitosan containing PTX [PTX-SHEC]) and evaluated its safety and efficiency using in vivo and in vitro experiments.</p><p><b>Methods:</b>The SHEC conjugated with PTX was confirmed by nuclear magnetic resonance (NMR) and Fourier-transform infrared spectroscopy (FT-IR) measurements. Drug releases in vitro and in vivo were determined using high-performance liquid chromatography. Cell viability in vitro was measured using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide. Rabbit biliary stricture model was constructed. All rabbits randomly divided into five groups (n = 8 in each group): the sham-operated rabbits were used as control (Group A), Groups B received laparotomies and suture, Group C received laparotomies and covered SHEC suture without the PTX coating, Group D received laparotomies and covered PTX-SHEC suture, and Group E received laparotomies and 1000 μmol/L PTX administration. Liver function tests and residual dosage of PTX from each group were measured by enzyme-linked immunosorbent assay. Histological data and α-smooth muscle actin (SMA) immunohistochemical staining of common bile duct were examined.</p><p><b>Results:</b>NMR and FT-IR indicated that PTX was successfully introduced, based on the appearance of signals at 7.41-7.99 ppm, 1.50 ppm, and 1.03 ppm, due to the presence of aromatic protons, methylene protons, and methyl protons of PTX, respectively. No bile leak was observed. The PTX-conjugated film could slowly release PTX for 4 weeks (8.89 ± 0.03 μg at day 30). The in vitro cell viability test revealed significantly different levels of toxicity between films with and without PTX (111.7 ± 4.0% vs. 68.1 ± 6.0%, P < 0.001), whereas no statistically significant difference was observed among the three sets of PTX-contained films (67.7 ± 5.4%, 67.2 ± 3.4%, and 59.1 ± 6.0%, P > 0.05). Histological examinations revealed that after 28 days of implantment, Groups D and E (but not Group C) had less granulation tissue and glandular hyperplasia in the site of biliary duct injury than Group B. The pattern was more obvious in Group D than Group E. Less α-SMA-positive cells were found in tissue from Groups D and E. Comparing with Group E, the liver function was improved significantly in Group D, including total bilirubin (2.69 ± 1.03 μmol/L vs. 0.81 ± 0.54 μmol/L, P = 0.014), alanine aminotransferase (87.13 ± 17.51 U/L vs. 42.12 ± 15.76 U/L, P = 0.012), and alkaline phosphatase (60.61 ± 12.31 U/L vs. 40.59 ± 8.78 U/L, P < 0.001).</p><p><b>Conclusions</b>PTX-SHEC film effectively inhibites the myofibroblast proliferation and extracellular matrix over-deposition during the healing process of biliary reconstruction. This original film might offer a new way for reducing the occurrence of the benign biliary stricture.</p>

ABSTRACT

Objective To find out the cut-off value of rose bengal plate agglutination test(RBPT) in rapid diagnosis of brucellosis.Methods From May to June 2009,398 people who came to the outpatient department of Inner Mongolia Center for Endemic Disease Control and Research were diagnosed brucellosis by RBPT and tube agglutination test (SAT).Tube agglutination test as a gold standard,rose bengal plate agglutination test,its authenticity and reliability were evaluated.Results Taking positive predictive value 100.0% as the selection standard,the cut-off value of RBPT was ++ which could be used to diagnose brucellosis.The sensitivity was 83.3%; the specificity was 100.0%; the Youden index was 0.832; and the compliance rote was 89.9%.Conclusion The cut-off value ++ of RBPT to diagnosis brucellosis is worthy of clinical promotion.

ABSTRACT

<p><b>OBJECTIVE</b>To probe an effective therapeutic method for the treatment of shoulder periarthritis.</p><p><b>METHODS</b>Ninety-six patients were randomly divided into an acupuncture group and an abdominal acupuncture group, 48 cases in each. According to clinical pathological staging, the disease was divided into pain stage and adhesion stage. In abdominal acupuncture group, 24 cases were in pain stage and treated with acupuncture at Zhongwan (CV 12), Shangqu (KI 17) on the healthy side, Huaroumen (ST 24) on the affected side and others; 24 cases were in adhesion stage and treated with acupuncture at Zhongwan (CV 12), Xiawan (CV 10), Qihai (CV 6), Guanyuan (CV 4) and others. In acupuncture group, there were 24 cases in either pain stage or adhesion stage, treated with acupuncture at Jianyu (LI 15), Jianliao (TE 14), Jianzhen (SI 9) and others on the affected side. In either group, the treatment was given three times a week, three weeks treatment made one session, 9 treatments totally. Before and after treatment, the Visual Analogue Scale (VAS) was adopted for pain assessment and functional activity score (Melle score) was for shoulder joint function assessment for all the patients.</p><p><b>RESULTS</b>After one session treatment, the efficacy was compared between two groups. In abdominal acupuncture group, the total effective rates were 95.8% (23/24) and 91.7% (22/24) in pain stage and adhesion stage respectively. In acupuncture group, the total effective rates were 91.7% (22/24) and 79.2% (19/24) in pain stage and adhesion stage respectively. The efficacy in adhesion stage of abdominal acupuncture group was better than that in acupuncture group (P < 0.05). After treatment, VAS score and Melle score were all reduced as compared with those before treatment in either group (all P < 0.01), in which, the reducing ranges in abdominal acupuncture group were larger than those in acupuncture group (all P < 0.05).</p><p><b>CONCLUSION</b>Abdominal acupuncture achieves a significant efficacy on shoulder periarthritis in stages. It is an effective therapy for shoulder periarthritis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the clinical curative effect of the new Bian-stone therapy.</p><p><b>METHODS</b>According to clinical trail principle of multi-centers, randomized grouping, parallel control and single-blind, the observation group (n=120) were treated by the new Bian-stone therapy, with multi-functional Bian-stone respectively dredging qi and blood of channels on the neck and shoulder, the shoulder and back, the limbs, and the control group (n=120) by electroacupuncture with Jianyu (LI 15), Jianliao (SJ 14), Jianzhen (SI 9), and other points selected. Their therapeutic effects were compared.</p><p><b>RESULTS</b>The two therapies had effects of analgesia, improving functional activity and transient analgesia. The total effective rate of the analgesic effect was 86.8% in the observation group and 72.5% in the control group with a significant difference between the two groups (P < 0.05), the observation group being better than the control group; the total effective rate of shoulder functional activity was 86.8% in the observation group and 79.8% in the control group with no significant difference between the two groups (P > 0.05).</p><p><b>CONCLUSION</b>The new Bian-stone therapy has obvious therapeutic effect on scapulohumeral periarthritis, with effects of analgesia, improving functional activity, and transient analgesia, and good long-term therapeutic effect.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the basic therapeutic function of Tiaokou (ST 38).</p><p><b>METHODS</b>According to clinically multi-central randomized controlled and single-blind test principle, 257 cases of periarthritis of shoulder were divided into two groups, a test group (n = 124) treated with oral anti-inflammatory analgesic medicine combined with acupuncture at Tiaokou (ST 38), and a control group (n = 133) treated with oral anti-inflammatory analgesic medicine. Their therapeutic effects were compared.</p><p><b>RESULTS</b>The total effective rate for stopping pain was 96.0% in the test group and 91.7% in the control group with a very significant difference between the two groups (P< 0.01). And the total effective rate for improvement of shoulder activity was 86.3% in the test group and 59.4% in the control group with a very significant difference between the two groups (P<0.01).</p><p><b>CONCLUSION</b>Oral anti-inflammatory analgesic medicine combined with acupuncture has obvious therapeutic effect on periarthritis of shoulder, which is better than that of simple oral anti-inflammatory analgesic medicine.</p>

ABSTRACT

Objective To explore the microbial etiology of early onset pulmonary infection after liver transplantation.Methods The 40 episodes of early onset pulmonary infection in 75 patients receiving liver transplantation in our hospital were reviewed and analyzed retrospectively.Results The incidence rate of pulmonary infection was 53.30% in these patients.A total of 54 pathogens were isolated,including 41(75.9%)strains of bacteria and 13(24.1%)fungal isolates.Gram-negative bacillus ac- counted for 68.3%(n=28)of the bacterial isolates,11(39.3%)of which were positive for extended spectrum?-lactamase. Gram-positive coccus accounted for 31.7%(n=13).Seven (58.3%)of the staphylococcal strains were methicillin-resistant. Conclusions The incidence of pulmonary infection is high early af- ter liver transplantation.Most of the isolated pathogens are drug resistant.Culture of sputum or secretion of lower respiratory tract and bedside X-ray examination are useful for the diagnosis.

ABSTRACT

<p><b>OBJECTIVE</b>To introduce Meta-analysis in evaluating diagnostic tests.</p><p><b>METHOD</b>Adjusted SROC method was used in assessing 7 diagnostic tests on fatty liver.</p><p><b>RESULTS</b>The pooled sensitivity and specificity of type B ultrasonography were 0.89 [95% confidence interval (CI): 0.87-0.92] and 0.94 (95% CI: 0.92-0.96) respectively while the Q value was 0.90 by adjusted SROC method. The results indicated that the diagnostic value of type B ultrasonography were high, thus could be regarded as an effective method for fatty liver diagnosis.</p><p><b>CONCLUSION</b>Meta-analysis on evaluating diagnostic tests could be used to assess the diagnostic test to increase the power of conclusion, and to improves its reliability.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To systematically assess imaging diagnostic tests for fatty liver and provide a decision-making basis for clinical diagnosis and screening.</p><p><b>METHODS</b>Electronic searches were conducted on the Chinese Biomedical Database, PubMed, and EMBASE, combining with manually searching of Chinese literature. All searches were completed until November 2002. All studies which evaluated imaging diagnostic test of human fatty liver were included. Data of diagnostic accuracy in the included studies were extracted, and methodological quality of the studies was assessed independently by two reviewers according to the established quality standard. Quantitative analysis or qualitative description were performed based on available data.</p><p><b>RESULTS</b>Of 13 studies that met the eligibility criteria, 10 studies evaluated the diagnostic accuracy of B-mode ultrasound, 3 studies evaluated contrast-enhanced (helical) CT. To assess 7 diagnostic test studies for fatty liver that used liver biopsy as reference test: the pooled sensitivity of B-mode ultrasound was 0.89 (95% confidence interval 0.87-0.92), specificity was 0.94 (95% confidence interval 0.92-0.96) and the Q value was 0.90 by adjusted SROC method. To assess 2 diagnostic test studies for fatty liver that used CT as reference test: the pooled sensitivity, specificity, and Q value were 0.92 (95% confidence interval 0.89-0.96), 0.88 (95% confidence interval 0.84-0.92), and 0.90 respectively by adjusted SROC method.</p><p><b>CONCLUSIONS</b>B-mode ultrasound method can be regarded as an effective method for fatty liver diagnosis and screening. The methodologic quality of diagnostic test needs to be improved.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the risk of stomach cancer associated with cigarette smoking.</p><p><b>METHODS</b>Meta-analysis was used to make formal summary statements about the relationship between smoking and stomach cancer. The results of the studies were analyzed by studied type(case-control study and cohort study) and gender(male and female). Homogeneity showed that random effect model should be selected to calculate the pooling relative risk (RR) and its corresponding 95% confidence interval (95%CI).</p><p><b>RESULTS</b>The total pooling RR(95%CI) was 2.14(1.12-4.06), while the total pooling RRs (95%CI) of male and female smoking with stomach cancer were 1.70 (1.34-2.14) and 1.83 (0.10-33.09), respectively.</p><p><b>CONCLUSIONS</b>Cigarette smoking is a risk factor for stomach cancer. Male smoking can increase risk of stomach cancer, however, the relationship between female smoking and stomach cancer should be studied further.</p>
